اثر پیش تیمار دمایی، نور و تنظیم کننده های رشد گیاهی بر القاء کالوس در کشت بساک کدوی پوست کاغذی (Cucurbita pepo var. Styriaca)

چکیده

این مطالعه به منظور بررسی اثر فاکتورهای مختلف بر القاء کالوس و تولید رویان هاپلوئید در کشت بساک گیاه دارویی کدوی پوست کاغذی (Cucurbita pepo var. Styriaca) انجام شد. برای انجام آزمایشات، بذور کدوی پوست کاغذی در گلخانه کشت و در زمان گلدهی، غنچه­ های نر با اندازه 5/1 سانتی­متر انتخاب و برای استفاده در مراحل بعدی استریل شدند. آزمایشات به صورت فاکتوریل در قالب طرح کاملاً تصادفی با 3 تکرار اجرا گردیدند. آزمایش اول اثر پیش تیمار سرمایی در سه سطح پیش تیمار سرمایی غنچه، پیش تیمار سرمایی بساک و بدون پیش تیمار سرمایی (شاهد) و ترکیب تنظیم کننده­ های رشدگیاهی بر صفت میزان کالوس­ زایی در کشت بساک مورد مطالعه قرار گرفت . پیش تیمار سرمایی غنچه ها و کشت بساکها در محیط کشت حاوی mgl-11 NAA و  mgl-1 2 2,4-D بیشترین درصد کالوس ­زایی را ایجاد کرد. در آزمایش دوم، اثر پیش تیمار حرارتی بر صفت درصد کالوس ­زایی بررسی شد. در این آزمایش بهترین جواب مربوط به  تیمار ºC32 به مدت 7 روز بود. آزمایش سوم شامل اثر تیمار نوری وتنظیم کننده­ هاِی رشد بر صفت کالوس ­زایی در کشت بساک کدوی پوست کاغذی بود، بساک­هایی که در شرایط نور با غلظت هورمونی mgl-1 5/2 2,4-D +  mgl-1 1 BAP کشت شد، بالاترین درصد کالوس­ زایی را نشان داد.  در آزمایش چهارم آندروژنز از طریق کشت میکروسپور در محیط کشت E20 اعمال گردید که منجر به رشد تعدادی از میکروسپورها شد و در  بررسی های سیتولوژیکی  تقسیمات قرینه مشاهده شد.

چکیده [English]

This study was performed to investigate the effect of various factors on callus induction and haploid embryo production in the anther culture of Pumpkin (Cucurbita pepo var. Styriaca). For experiments, pumpkin seeds were planted in the greenhouse and at the time of flowering, male flower buds with a size of 1.5 cm were selected and sterilized for use in later stages. The experiments were performed as a factorial experiment in a completely randomized design with 3 replications. The first experiment studied the effect of cold pretreatment on three levels of bud cold pretreatment, anther cold pretreatment and without cold pretreatment (control) and the combination of plant growth regulators on callus formation in anther culture. Cold pretreatment of buds and culture of anthers in culture medium containing 1 mgl-1 NAA and 2 mgl-1 2,4-D produced the highest callus formation percentage. In the second experiment, the effect of heat pretreatment on the percentage of callus formation was investigated. In this experiment, the best response was related to 32°C treatment for 7 days. The third experiment consisted of the interaction effectt of light treatments and growth regulators on callus formation in anther culture of Pumpkin. Anthers that cultured in light conditions with a hormonal concentration of 2.5 mgl-1 2,4-D + 1 mg /l BAP showing the highest percentage of callus formation. In the fourth experiment, androgenesis was applied by culturing microspores in E20 medium, which led to the growth of a number of microspores, and in the cytological examination, the symmetric divisions were observed.
